بهینه‌سازی شرایط عملیاتی استخراج اسیدهای آمینه با کربن‌دی‌اکسید فوق بحرانی

چکیده
هدف از این تحقیق تعیین شرایط بهینه برای استخراج گزینش ­پذیر آمینواسید­های آزاد بهروش استخراج فوق بحرانی است. دو متغیر دما، فشار با انتخاب زمان ثابت 60 دقیقه در این راستا و بااستفادهاز سامانۀ طراحی آزمایش یک متغیر در یک زمان، مطالعه شد. در گام اول مشخص شد که شگرد استخراج فوق بحرانی صرفاً نسبتبه پنج آمینواسید آلانین، گلایسین، سرین، هیستیدین و گلوتامیک اسید استخراج گزینش ­پذیر داشته و نسبتبه سایر آمینواسیدها در بازه عملیاتی مورد مطالعه، عملکرد استخراجی ضعیفی دارد. پس از بهدست‌آمدن داده­ های تجربی، در نرمافزار اکسل مدل کرداری داده­ ها تهیه شد، که نشاندهندۀ بالاترین سطح گزینش ­پذیری و بازده استخراج برای گلوتامیک­ اسید در فشار 14 مگاپاسکال و دمای 20 درجۀ سلسیوس، برای گلایسین در فشار 16 مگاپاسکال و دمای 60 درجۀ سلسیوس و برای آلانین، سرین و هیستیدین در فشار20 مگاپاسکال و دمای 60 درجۀ سلسیوس بود. بالاترین بازده استخراج (5/3%) در شرایط بهینۀ استخراج گلوتامیک ­اسید است که بالاترین درصد در نمونه را دارد.

چکیده English

This research aimed to investigate optimum operating condition for selective extraction of free amino acids by supercritical carbon dioxide (SFE). OVAT experiment design technique is used, setting temperature and pressure as variables and setting time at 60 minute as constant. Results show that SFE has good efficiency in extraction of alanine, glycine, serine, glutamic acid and histidine while showing unsatisfactory results on others under the chosen operating conditions for this study. Experimental extraction results are plots in excel software which shows maximum selectivity and yield for glutamic acids at (P=14 Mpa T=20 °C) and for glycine at (P=16 Mpa T=60 °C) and for alanine, serine and histidine at (P=20 Mpa T=60 °C). The highest extraction yield (3.5%) was achieved at conditions where glutamic acid, the most abundant amino acid in the sample, had the highest exaction yield.
